IGCSE Biology: Biological Molecules Practice Questions

Carbohydrates, fats and proteins are the three main biological molecules. Carbohydrates and fats contain carbon, hydrogen and oxygen, while proteins also contain nitrogen. Each has a standard chemical test with a specific colour change.

Topic 4 of Cambridge IGCSE Biology 0610 is largely about food tests, and food test questions are marked strictly. You need the reagent, the procedure including any heating, and the colour change stated from and to. Vague answers such as it turns positive score nothing. Question 1[4 marks]

Describe how you would test a food sample for reducing sugar, and state the result if reducing sugar is present.

Answer: Add Benedict's solution and heat in a water bath. The blue solution turns orange or brick red.
  1. Grind the food and mix it with distilled water to make a solution, then filter it.
  2. Add an equal volume of Benedict's solution to a sample in a test tube.
  3. Heat the tube in a water bath at about 80 degrees Celsius for a few minutes. Heating is essential, since the reaction does not occur at room temperature.
  4. A positive result changes the blue solution through green and yellow to orange or brick red. If no reducing sugar is present it stays blue.
How the marks are awarded. 1 mark for adding Benedict's solution. 1 mark for heating in a water bath. 1 mark for the starting colour blue. 1 mark for the final colour orange, red or brick red.
Where students lose the mark. Writing only that it turns red. The mark scheme wants the change stated from blue to orange or red, because a colour change means both ends of it.
Question 2[4 marks]

A student has one sample that contains starch and protein but no fat. Describe the two tests she would carry out and the results she would expect for each.

Answer: Iodine turns blue black for starch. Biuret turns purple for protein.
  1. Test 1: add a few drops of iodine solution to the sample. It changes from orange brown to blue black, showing starch is present.
  2. Test 2: add biuret solution to a fresh sample. It changes from blue to purple or lilac, showing protein is present. No heating is needed.
  3. If she also tested for fat with the ethanol emulsion test, the mixture would remain clear rather than turning cloudy white.
  4. Each test must be carried out on a separate sample so that the reagents do not interfere with one another.
How the marks are awarded. 1 mark for iodine solution. 1 mark for blue black. 1 mark for biuret solution. 1 mark for purple or lilac.
Where students lose the mark. Using the same test tube for both tests. Reagent colours mask each other, so each test needs a fresh sample.
Question 3[3 marks]

DNA consists of two strands coiled together. Explain how the two strands are held together and state the base pairing rule.

Answer: Cross links between pairs of bases hold the strands together. A pairs with T, and C pairs with G.
  1. The two strands are coiled around each other to form a double helix.
  2. They are held together by cross links formed between bases on opposite strands.
  3. Adenine always pairs with thymine.
  4. Cytosine always pairs with guanine. This means the sequence on one strand determines the sequence on the other.
How the marks are awarded. 1 mark for cross links between bases holding the strands together. 1 mark for adenine pairing with thymine. 1 mark for cytosine pairing with guanine.
Where students lose the mark. Pairing adenine with cytosine. Learn the pairs as A with T and C with G, and note that the letter shapes in each pair are dissimilar.
Question 4[3 marks]

Name the elements present in a protein molecule and explain how this differs from a carbohydrate.

Answer: Proteins contain carbon, hydrogen, oxygen and nitrogen. Carbohydrates contain carbon, hydrogen and oxygen only.
  1. Proteins contain carbon, hydrogen, oxygen and nitrogen. Some proteins also contain sulfur.
  2. Carbohydrates contain only carbon, hydrogen and oxygen.
  3. The difference is therefore the presence of nitrogen in proteins.
  4. This is why nitrate ions are needed by plants to make amino acids, and therefore proteins.
How the marks are awarded. 1 mark for carbon, hydrogen and oxygen in protein. 1 mark for nitrogen in protein. 1 mark for identifying nitrogen as the difference, or for stating that carbohydrates contain only C, H and O.
Where students lose the mark. Listing nitrogen for carbohydrates. Only proteins among the three main food groups contain nitrogen.

Why does the Benedict's test need heating?

The reaction between Benedict's solution and a reducing sugar only proceeds at a raised temperature. Without heating in a water bath the solution stays blue even when reducing sugar is present, giving a false negative. Cambridge awards a specific mark for stating that the tube is heated.
